  • Macrophage cell culture: Conduct Cell Death Assay infected with various pathogens such as S. Typhi, S. Paratyphi A, S. Typhimurium and E. Coli
  • Protein expression and purification: The sequence of those S. Typhi candidate antigens cloned into 5’NcoI and 3’NotI restriction sites of pET28b for His-Tag purification with E. Coli BL21(DE3) pLysS for expression and purification soluble and insoluble recombinant protein
  • Enzyme-Linked ImmunoSorbent Assay: ELISAs employed to detect antigens specific antibodies class IgA, IgM and IgG with acute, convalescent, asymptomatic chronic and longitudinal immune response in ALS and human plasma typhoid fever that confirmed S. Typhi (ST), S. Paratyphi A (SPA) or other infections, undiagnosed febrile disease and negative control population
  • Immuno-strip for rapid diagnostic test: The candidate S. Typhi antigens were immobilized on a nitrocellulose membrane and subsequently tested with ST, SPA, and undiagnosed febrile plasma samples
  • Immuno-fluorescence staining assay: The expression of S. Typhi antigens was visualized, with images captured using a wide-field fluorescent microscope
  • Serum Bactericidal Assay (SBA): Mice sera, immunized to different specific antigens or clinical typhoid patient sera to kill wild-type or mutant S. Typhi and S. Paratyphi A strains
  • Knockout genes of interested: Homologous recombination with site-specific mutagenesis
  • Transposon Directed Insertion Sequencing-TraDIS: The mutant library with clinical S. Typhi and S. Paratyphi A isolates were generated by Tn5 transposon
  • Molecular biology: Techniques employed included DNA, RNA, Protein extraction, PCR, gene sequencing, flow cytometry, electroporation transformation, and SDS PAGE Microbiology: Microbiological cultures, and testing methodologies pertinent to research and diagnostic laboratories, as well as pharmaceutical manufacturing, were conducted  R and Python programming software: statistic, analysis, visualization, plot with meta data of Serology, SBA, TraDIS, Genome, OMIC and Cytokines, S. Typhi genes functional classification using KEGG pathways or Gen Ontology  Clinical Research Associate: collect, manage and analysis Case Report Form (CRF)  Typhoid Clinical Vaccine Trial: TyVAC study and STRATAA study
